RAD51 is one of the key factors of DNA repair by homologous recombination and has been shown to have anti-apoptotic activity in tumor cells. RAD51 protein interacts with a variety of tumor suppressor proteins including p53, BRCA1 and BRCA2. Elevated expression of RAD51 enhances radio-resistance of human tumor cells. Overexpression of RAD51 protein in tumor cells renders them resistant against cytotoxic drugs like Cisplatin. RAD51 interacts with BRCA1 and BRCA2 to influence subcellular localization and cellular response to DNA damage. BRCA2 inactivation may be a key event leading to genomic instability and tumorigenesis from deregulation of RAD51. High-level expression of RAD51 has been observed in a variety of human malignancies. Allograft Inflammatory Factor 1 (also known as AIF1 or IBA1) is a cytoplasmic, calcium-binding protein that is thought to play a role in macrophage activation and function. AIF1, containing two EF domains, is induced by cytokines and Interferons. In an unstimulated state, AIF1 colocalizes with actin, and upon stimulation, translocates to lamellipodia. It is also a marker of human microglia and is expressed by macrophages in injured skeletal muscle. The gene encoding AIF1 resides in the tumor necrosis factor (TNF) cluster of genes, located in the region represented by the human major histocompatibility complex (MHC). Complement C1q A Chain (C1qa) is a subunit of C1q. C1q is composed of 6 polypeptide chains each of C1qa, C1qb, and C1qc. C1q is a component of the classical complement pathway, and thereby plays an important role in the immune response. Each C1q subunit contains an N-terminal collagen-like region and a C-terminal C1q globular domain. The presence of receptors for C1q on effector cells modulates its activity, which may be antibody-dependent or independent. Macrophages are the primary source of C1q, while anti-inflammatory drugs as well as cytokines differentially regulate expression of the mRNA as well as the protein. This MAb recognizes a protein of 52 kDa, which is identified as TNFR1 (TNFR-associated factor 1). CD30-positive lymphoproliferations of the skin comprise 30% of all primary cutaneous T-cell lymphomas (CTCLs). Besides borderline cases this group includes lymphomatoid papulosis (LyP) and primary cutaneous anaplastic large T-cell lymphoma (cALCL). Although the two entities overlap clinically, histopathologically, immunopathologically and genetically, they differ considerably in their prognosis. In particular, common feature of both cases is histologically the presence of atypical lymphoid CD30-positive T blasts and genetically a clonal T-cell-receptor rearrangement. However, both cases differ considerably in their clinical course: Lesions of LyP regress spontaneously, whereas those of cALCL persist and may progress and spread. The metal-responsive element (MRE)-binding transcription factor (MTF-1) stimulates the expression of metallothioneins in response to the exposure of cells to heavy metals. MTF-1 contains six zinc fingers in the DNA binding domain. The phosphorylation of MTF-1 in response to metal exposure appears to play a significant role in the ability of MTF-1 to activate metallothionein transcription. In addition to its role in metallothionein activation, MTF-1 is involved in a post-transcription regulatory complex for ribosomal protein S25. MTF-1, La and p53 inhibit the nuclear export of S25 mRNA in response to nutrient depravation. Focal adhesions are identified as areas within the plasma membrane of tissue culture cells that adhere tightly to the underlying substrate. In vivo, these regions are involved in the adhesion of cells to the extracellular matrix. Paxillin and vinculin are cytoskeletal, focal adhesion proteins that are components of a protein complex which links the Actin network to the plasma membrane. Vinculin binding sites have been identified on other cytoskeletal proteins, including talin and alpha-actinin each contain actin binding sites. Reportedly, vinculin is a potential plasma biomarker for Age-related Macular Degeneration (AMD). Cytotoxic T lymphocytes (CTLs) recognize melanoma-associated antigens, which belong to three main groups. These groups include tumor-associated testis-specific antigens, melanocyte differentiation antigens and mutated or aberrantly expressed antigens, which are routinely used as markers to identify melanomas based on their binding to specific monoclonal antibodies. gp100, also designated ME20-M, ME20-S and PMEL 17, is classified as a melanocyte differentiation antigen and is expressed at low levels in normal cell lines and tissues, but is upregulated in melanocytes. gp100 is a highly glycosylated protein. This antibody recognizes a protein doublet of 20-22 kDa, identified as MART-1 (Melanoma Antigen Recognized by T cells 1) or Melan-A. MART-1 is a newly identified melanocyte differentiation antigen recognized by autologous cytotoxic T lymphocytes. Seven other melanoma associated antigens recognized by autologous cytotoxic T cells include MAGE-1, MAGE-3, tyrosinase, gp100, gp75, BAGE-1, and GAGE-1. Subcellular fractionation shows that MART-1 is present in melanosomes and endoplasmic reticulum. This MAb labels melanomas and other tumors showing melanocytic differentiation. It is also a useful positive-marker for angiomyolipomas. This MAb recognizes a protein of ~30 kDa, identified as CELA3B (Chymotrypsin like elastase family member 3B). Elastases form a subfamily of serine proteases that hydrolyze many proteins in addition to elastin. Humans have six elastase genes which encode the structurally similar proteins elastase 1, 2, 2A, 2B, 3A, and 3B. Unlike other elastases, elastase 3B has little elastolytic activity. Like most of the human elastases, elastase 3B is secreted from the pancreas as a zymogen and, like other serine proteases such as trypsin, chymotrypsin and kallikrein; it has a digestive function in the intestine. Elastase 3B preferentially cleaves proteins after alanine residues. Elastase 3B may also function in the intestinal transport and metabolism of cholesterol.
